Yes, it's definitely possible. The following code snippet does the show:

listen [::]:80 default ipv6only=on;
location / { proxy_pass http://127.0.0.1:8081; }
}

server {
listen 127.0.0.1:8081;
location / { return 200 "OK, 127.0.0.1:8081\n"; }
}


% curl 127.0.0.1:8081
OK, 127.0.0.1:8081
% curl -g -6 "http://[::1]/"
OK, 127.0.0.1:8081
